التزم يعقوب الحياة النسكية منذ نعومة أظفاره. وقد ترهّب في دير ستوديون في القسطنطينية وتتلمذ للقديس ثيودوروس المعيد له في 11 تشرين الثاني. وعن ثيودوروس أخذ يعقوب محبته الحارة لله وغيرته على الإيمان القويم. ويبدو أنه لمع في سيرة الفضيلة. وإذ لوحظت مزاياه الفريدة تمّ اختياره للأسقفية.
كأسقف عانى يعقوب الاضطهاد والنفي لأنه رفع لواء الأيقونات المقدسة ودافع عنها فيما كان مكرمو الأيقونات عرضة للملاحقة والتنكيل والأيقونات للتحطيم والإتلاف. وقد ورد ان يعقوب كابد الجوع والعطش وسوء معاملة العمال الذين نشرهم الأمبراطور، في ذلك الحين، تنفيذا لسياسته بشأن التخلص من الإيقونات وإكرامها وإخراس مشايعيها. أخيرا قضى ثابتا على الإيمان القويم متمسكا بتقليدات الآباء فأُحصي في عداد القديسين المعترفين. القديس ثيودوروس الستوديتي أكبر أمانته وغيرته في واحدة من رسائله.
تعيد له الكنيسة في 21 آذار.
